腹腔途径时脂质体和糖化多聚赖氨酸对外源基因肝脏靶向导入性能的比较 被引量:2

摘要 目的 比较脂质体和糖化多聚赖氨酸经腹腔途径时对目的基因肝脏靶向定位效果的影响。方法 将真核细胞表达质粒分别与脂质体和糖化多聚赖氨酸偶联 ,经腹腔注射导入大鼠体内 ,通过原位杂交和免疫组化的方法观察目的基因在肝脏和其他脏器的分布表达。结果 经脂质体或糖化多聚赖氨酸包埋的质粒DNA导入体内2 4h后均见明显表达 ,1周后逐渐下降 ,3周时仍有表达 ;均以肝脏为主要分布器官 ,但用糖化多聚赖氨酸包埋的质粒在肝脏中的分布表达高于使用脂质体的包埋 ,在其他脏器中的分布表达低于使用脂质体的包埋。结论 腹腔途径时糖化多聚赖氨酸对DNA的肝脏靶向定位效果优于脂质体。 Purpose To compare the effects of liposomes and glyco poly L lysine(G?PLL)peritoneally injected on targeted uptake and gene expression of liver. Methods After encapsulated by lipofectamine or galactose terminal glyco poly L lysine, the plasmid which can be expressed in eukaryotic cells was transferred into rat's body intraperitoneally, then the results of distribution and expression of the plasmid in liver and other organs were observed by in situ hybridization and immunohistochemistry. Results Twenty four hours after transferred in vivo the expression of the plasmid of the plasmid encapsulated by liposomes or G?PLL was obvious,one week later the expression began to decrease, and could still be found three weeks later.Both liposomes and G?PLL took the liver as the major distribution tissure.The distribution and expression of the plasmid bound to G?PLL was higher in liver than that encapsulated by liposomes and lower in other tissue than that encapsulated by liposomes.
