摘要
【目的】探讨二烯丙基三硫(DATS)抑制人卵巢癌CoCl增殖的作用。【方法】台盼蓝染色细胞计数法测定DATs对体外培养人卵巢癌CoCl细胞增殖的影响;软琼脂克隆形成法测定DATs对体外培养CoCl细胞的非锚定依赖性生长作用;PI染色流式细胞术(FCM)分析DATS对CoCl细胞周期的影响。【结果】细胞计数法结果显示DATs抑制体外培养人卵巢癌CoCl增殖和生长,呈剂量依赖性;软琼脂克隆形成法结果显示:DATS呈剂量依赖性抑制细胞集落形成;PI染色FCM结果显示不同浓度的DATS作用于CoCl细胞后,使细胞周期阻滞于G1期。【结论】DATS具有抑制卵巢癌CoCl生长的作用。
[Objective] To investigate the effect of diallyl tfisulfide(DATS) for inhibiting the proliferation of human ovarian cancer cell line CoC1 cells. [Methods] Trypan blue exclusion method was used to study the effect of DATS on the proliferation of human ovarian cancer CoC1 cells cultured in vitro. Soft agar colony formation test was used to study the effect of DATS on the non-anchoring dependent growth of CoC1 cells cultured in vitro. The effect of DATS on the cycle of CoC1 cells was analyzed by using PI-staining flow cytometry(FCM). [Results]The trypan blue exclusion method showed that DATS inhibited the proliferation and growth of CoC1 cells in a dose-dependent manner. Soft agar colony formation test showed that DATS inhibited the cell colony formation in a dose-dependent manner. H-staining FCM showed that different concentration of DATS for treating CoC1 cells arrested the cell cycle in G1 phase. [Conclusion] DATS can inhibit the growth of human ovarian cancer CoC1 cells.
