银杏叶提取物对大鼠脑出血后早期血肿周围神经元凋亡的作用

Effects of Ginkgo Biloba Extract on Apoptotic Neurons Around the Early Hematoma after Intracerebral Hemorrhage in Rats

目的探讨银杏叶提取物对大鼠脑出血后早期血肿周围神经元凋亡的影响。方法将36只SD大鼠随机分为正常组、出血组、术前24 h预防用药组、术后0 h治疗组、6 h治疗组和24 h治疗组,每组6只,出血组和治疗组采用尾状核注入Ⅶ胶原酶建立脑出血模型,预防用药组和各治疗组于相应时间点给予银杏叶注射液2 mL.kg-1腹腔注射,各组大鼠均于术后72 h处死。用免疫组化方法观察各组出血灶周围凋亡神经元的Bcl-2、Bax和Caspase-3的阳性细胞数量变化。结果 HE染色显示出血组大鼠尾状核部位的脑组织大量坏死,各治疗组大鼠尾壳核部位的出血坏死灶较出血组缩小,脑组织水肿减轻,其中术前24h预防用药组表现最为明显;Bcl-2、Bax和Caspase-3蛋白在正常组均有少量表达,脑出血后明显增加;预防用药组和各治疗组中,Bcl-2蛋白阳性细胞数较出血组明显升高(P<0.01),Bax与Caspase-3蛋白的阳性细胞数较出血组明显降低(P<0.01);预防用药组Bcl-2蛋白阳性细胞数比各术后治疗组多,Bax与Caspase-3蛋白的阳性细胞数比术后各治疗组少。结论银杏叶提取物能保护大鼠脑出血后血肿周围神经元,抑制神经元凋亡,并且早期用药效果较好。

Objective To observe early period apoptosis of nerve cells around the hematoma after intracerebral hemorrhage in rats and to discuss effect of Ginkgo biloba extract on the nerve cells around the hematoma.Methods 36 male SD rats were randomly divided into model group and normal group,24h treatment groups before surgery,0 h after the treatment group,6h treatment group,24h treatment group.All rats were killed at 72h after operation.Apoptosis was observed around the hematoma by immunohistochemistry.Results HE staining showed that rat caudate nucleus hemorrhage parts had extensive tissue necrosis;the treatment group had narrower site of rat caudate nucleus hemorrhage,necrosis.Bcl-2,Bax and Caspase-3 protein expression were mild in the normal group and significantly increased in ICH;Bcl-2 protein positive cells in preventive treatment group and the treated group were significantly higher than those in the ICH group（P0.01）;Bax and Caspase-3 protein positive cells in preventive treatment group and the treated group was significantly lower than those in ICH group（P0.01）;Bcl-2 protein positive cells in preventive treatment group were more than those in the postoperative treatment group and Bax and Caspase-3 protein cells in preventive treatment group and the treated group was less than those in postoperative treatment groups.Conclusion Ginkgo biloba extract can protect nerve cells around the hematoma after intracerebral hemorrhage in rats,inhibit neuronal apoptosis.Preventive medicine showed better results.
