摘要
通过改进豌豆核DNA的分离纯化方法,简化了分离纯化豌豆核DNA的操作程序;纯化了的G2 豌豆核DNA 的紫外吸收值A260/ A280 = 1 .92 ,Alaska 豌豆核DNA的紫外吸收A260/ A280 = 1 .88, 表明该方法提取的豌豆核DNA是高纯度的DNA,根据光吸收值测定产量收率,从每克豌豆幼苗中可获得约130 μg 核DNA,这为豌豆核DNA 的有关分子生物学研究提供了简便、经济、有效的分离纯化方法.
An improved method of separation and purification of nuclear DNA from pea tissue is described in this paper, which simplifies working processes. A 260 /A 280 of the purified nuclear DNAs of G 2 pea and Alaska pea are 1.92 and 1.88 respectively, these are typical value of absorption ultraviolet spectrum of nucleic acieds. It clearly showed that purified nuclear DNA are pure DNA. The output of nuclear DNA is about 130 micrograms per gram of seedling. This method is simple, economic and useful for further study on the nuclear DNA of pea.
