摘要
将人纤溶酶原kringle5功能区基因插入融合基因表达载体pET17bBamHⅠ位点,构建成重组质粒pETK23,转化大肠杆菌BL21(DE3),在IPTG诱导下,纤溶酶原kringle5功能区基因在重组转化菌株BL21(DE3,pETK23)中获得高效表达,表达产物以可溶性蛋白形式存在,表达量占菌体总蛋白的528%.
Kringle 5 of human plasminogen is a recently discovered inhibitor of endothelial cell growth and migration It may also supress angiogenesis and tumor growth The gene encoding kringle 5 of human plasminogen was cloned in the Bam HⅠsite of pET 17b giving rise to the plasmid pETK23 which was then transformed into E coli BL21(DE3) Kringle 5 of human plasminogen was efficently expressed in recombinant strain E coli BL21(DE3,pETK23) by IPTG induction The expression product constitutes about 52 8% of the total protein and exsists in the form of soluble protein Western blot analysis demonstrates that it reacts positively with goat anti human plasminogen antisera
