摘要
将亚硫酸盐制浆方法用作蔗渣原料预处理,研究了所得到的蔗渣纤维对纤维素酶吸附以及其水解糖化性能。利用分光光度法的动力学模式测定预处理后蔗渣对纤维素酶的初始吸附量,并讨论了蔗渣纤维对纤维素酶的吸附率与纤维水解率的关系。实验结果表明:亚硫酸盐预处理温度的升高和用药量增加可以提高蔗渣纤维对纤维素酶的初始吸附率;水解产生的葡萄糖和总糖得率随着纤维素酶初始吸附率的增加而增加;当纤维素酶在蔗渣纤维上的吸附率由4.04%升至46.77%时,总糖得率由15.58%增加到82.22%。纤维素酶在纤维上的吸附率达到37%后,蔗渣的酶水解明显增加。
A novel process by using bisulfite pulping method for bagasse pretreatment to improve enzyme adsorption on fibers and their enzymatic hydrolysis were studied.The cellulase adsorption on the pretreated bagasse was measured by spectrophotometry with a kinetic model,and then the relationship between cellulase adsorption and the yield of hydrolysis was discussed.The results show that the rising of temperature of sulfite pretreatment and increasing of the dosage can improve the cellulase adsorption on the pretreated bagasse.With the higher cellulase adsorption on the pretreated bagasse,the higher sugar hydrolysis yield(mainly glucose and xylose) are obtained.When cellulase adsorption on bagasse fiber increases from 4.04% to 46.77%,the sugar hydrolysis yield increases from 15.58% to 82.22%;when cellulase adsorption on the pretreated bagasse is over 37%,its hydrolysis yield increases remarkably.
