双花提取物抗糖尿病的作用研究

摘要 目的:探讨月季和玫瑰双花提取物对糖尿病糖脂代谢的影响。方法:以高脂饲料结合小剂量链脲佐菌素建立2型糖尿病大鼠模型,灌胃不同剂量双花提取物2周后进行口服糖耐量试验,5周后测血清胰岛素(Ins)、胰高血糖素(Glu)、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)、糖化血清蛋白(GSP)、空腹血糖(FBG),同时测心脏、肝脏、肾脏丙二醛(MDA)含量;利用肝癌细胞株HePG2观察双花提取物对葡萄糖消耗的影响。结果:双花提取物各剂量均可改善糖耐量,降低糖尿病大鼠FBG,TC,TG,LDL-C,GSP,Ins,Glu,MDA,升高HDL-C,以高剂量效果最佳;另外双花提取物可显著增加HePG2葡萄糖消耗量,并呈剂量相关性。结论:双花提取物可显著降低2型糖尿病大鼠血糖水平,调节血脂异常,其机制可能包括改善糖耐量及胰岛素抵抗,促进肝脏对糖的利用,对防治糖尿病及其并发症有积极作用。 Objective:The PurPose of this study was to investigate the effects of the comPound flower extract made of rosa chinensis and rosa rugosa on metabolism of glucose and liPid in tyPe II diabetic mellitus(DM)rats.Methods:In vivo,SD rats were fed with high fat diet and then given iP STZ to establish tyPe II diabetic mellitus rat model.Oral glucose tolerance test was conducted after ig medication for two weeks,and the contents of insulin(Ins),glucagon(Glu),total cholesterol(TC),triglyceride(TG),high-density liPoProtein cholesterol(HDL-C),low-density liPoProtein cholesterol(LDL-C),glycosylated serum Protein(GSP),fasting Plasma glucose(FBG)in serum were detected at the end of the exPeriment.Meanwhile,malondialdehyde(MDA)of several organs were also measured.In vitro,the effect of the extract on glucose consumPtion(GC)in HePG2 cells was tested.Results:In vivo,glucose tolerance of rats treated with different dose of the extract was imProved,the levels of FBG,TC,TG,LDL-C,GSP,Ins,Glu,MDA was also reduced and HDL-C enhanced comPared with that of model rats.The high dose of the extract showed its strong activity.In vitro,the GC treated with extract in HePG2 cells was increased in a concentration dePendent way.Conclusion:The comPound flower extract reduces Plasma glucose level of DM rats effectively,regulates hyPerliPidemia and its Potential mechanism may include imProving insulin resistance,enhancing GC of liver.This extract is benefit for relieveing diabetes and its comPlications.
