摘要
目的使用特定细胞因子刺激诱导分离扩增后的兔骨髓间充质干细胞(Mesenchynml stem cells,MSCs),在体外试验中使其分化为软骨细胞,为后续构建工程化半月板研究提供种子细胞,探讨兔MSCs重建半月板组织的可行性和有效性。方法用碱性成纤维细胞生长因子(Basic fibroblast growth factor,bFGF)和转化生长因子β1(Transforming growth factor-βl,TGF-β1)协同刺激已分离并经体外传代培养扩增的兔MSCs,通过倒置细胞显微镜观察及免疫组化检测,证明其已经向软骨细胞系分化。结果用bFGF和TGF-β1协同刺激已分离并经体外扩增的兔MSCs后,细胞生长速度增快,免疫组织化学检测提示向软骨细胞系分化。结论兔MSCs可向软骨细胞分化;bFGF和TGF-β1能加快MSCs的体外增殖并促使其向软骨细胞系分化,可为构建工程化半月板提供理想的自体来源种子细胞。
Objective To study whether rabbit bone marrow-derived mesenchymal stem cells(MSCs) can be induced to differentiate into the chondrocytes lineage with the stimulation of basic fibroblast growth factor(bFGF) and transforming growth factor-β1(TGF-β1) . Methods After stimulated,evidences were looked for to prove whether the MSCs had entered the chondrocyte 1ineage with inverted microscope and immunohistochemistry of collagen type I and type II. Results After stimulated,cell growth was speeded up,and collagen type I and type II were detected in the post-stimulated MSCs,which-can differentiate into chondrocyte lineage. Conclusion With the stimulation of bFGF nd TGF-β1,the MSCs can differentiate into the chondrocyte lineage,and autogenous seed cells can be provided by this method.
