杭白芍炮制前后特征图谱比较研究

Comparative Study on the Characteristic Chromatogram of the Crude and Processed Radix Paeoniae Alba from Hangzhou

目的采用HPLC法研究浙江产白芍炮制前后特征图谱的变化,为科学评价和控制其质量提供有效的方法。方法以70%甲醇为提取溶剂,采用加热回流的方法提取制备供试样品。以Hanbon C18色谱柱为固定相,乙腈-0.1%磷酸水溶液为流动相进行梯度洗脱,检测波长230 nm,柱温25℃,进样量20μL,流速1.0 mL/min,进行HPLC特征图谱检测。以芍药苷为对照品,检测药材炮制前后芍药苷含量变化。结果白芍经炮制后,其不同炮制品间共确定9个主要共有特征峰。研究发现特征图谱中主要色谱峰发生了显著改变,与炮制前相比峰面积显著降低。表明白芍经炮制处理后,主要化学成分含量明显减少。结论建立了一种重现性、精密性良好的HPLC特征图谱检测方法,可用于浙江产白芍及其炮制品的质量控制。

OBJECTIVE To establish a sensitive and specific HPLC method for quality control of Radix Paeoniae Alba.METHODS HPLC method was applied for quality assessment of Radix Paeoniae Alba.HPLC analysis was performed on a Hanbon C18 column（250 mm×4.6 mm,5μm）.The mobile phase consisted of acetonitrile（solvent B） and water containing 0.1%（v/v） phosphoric acid（solvent A） at a constant flow rate of 1.0mL/min.An increasing linear gradient（v/v） of solvent B was used.The injection volume was 20μL.The column temperature was set at 25℃.The chromatograms were monitored at 230nm.RESULTS The HPLC characteristic chromatogram of Radix Paeoniae Alba,showing 9 characteristic peaks,was established from its prepared samples of Radix Paeoniae Alba.CONCLUSION The characteristic chromatogram of Radix Paeoniae Alba with high specificity can be used to control its quality and assure lot-to-lot consistency.
