摘要
在不同时间、温度、浓度等条件下,利用紫外分光光度法研究氟罗沙星(FLRX)、洛美沙星(LMF)与鲑鱼精DNA的相互作用。结果表明:鲑鱼精DNA紫外吸收值分别随溶液中FLRX和LMF浓度的增加而增加;在pH4.0~11.0条件下,FLRX、LMF分别与DNA大分子以槽沟式结合。根据实验结果利用双倒数分析法计算出FLRX、LMF分别与DNA相互作用的结合常数,分别为KDNA-FLRX=0.223×104 L/mol,KDNA-LMF=0.290×104 L/mol。
The interaction, at different times, different temperatures and different concentrations, between FLRX,LMF and Salmon sperm DNA were studied by UV-visible spectrophotometry. The results showed that Salmon sperm DNA's UV adsorption values increas with the concentration of both quinolones increasing. The two quinolones bind DNA's groove at pH4. 0 - 11.0. The combination constants between FLRX,LMF and Salmon sperm DNA were calculated by double reciprocal analysis and the combination constants were KDNA-FLRX = 0. 223 ×10^4 L/tool and KDNA-LMF= 0. 290 × 10^4 L/mol.
