摘要
目的应用全自动活细胞观测分析系统(Cell-IQ)及流式细胞仪观察榄香烯诱导人胃癌细胞BGC-823凋亡的影响。方法应用不同浓度的榄香烯作用于人胃癌BGC-823细胞,使用Cell-IQ筛选抑制增殖的最佳浓度,并根据最佳作用浓度,应用流式细胞仪观察分析榄香烯对人胃癌BGC-823细胞凋亡的影响。结果榄香烯对人胃癌BGC-823细胞的抑制作用随其浓度增加而逐渐增强,以150μg/mL为最佳,之后榄香烯浓度再增高,其抑制肿瘤细胞增殖作用不再增强;150μg/mL的榄香烯作用于胃癌BGC-823细胞24 h后,主要表现为诱导早期凋亡为主,细胞凋亡率为(36.9±3.23)%,较对照组有明显差异(P<0.01)。结论 Cell-IQ能够进行细胞毒性、活性和增殖的检测和筛选;榄香烯对于人胃癌BGC-823细胞有明确抑制作用,最佳浓度为150μg/mL;榄香烯抑制人胃癌BGC-823细胞增殖作用的主要机制可能为诱导细胞凋亡,而非细胞毒作用。
Objective To observe the effect of elemene on inducing the apoptosis of gastric cancer cells BGC-823 with the help of Cell-IQ and flow cytometry.Methods We used elemene of different concentration to affect the BGC-823,and then Cell-IQ was used to select the optimum concentration.According to the optimum concentration,we observed and analyzed the influence of elemene on apoptosis with the help of flow cytometry.Results The effect of inhibiting gastric cancer cell BGC-823 proliferation was better when the concentration of elemene was higher with the optimum concentration of 150 μg/mL.However,the concentration of elemene higher than 150 μg/mL could not improve its effects on inhibiting tumor cell growth.Twenty-four hours after the elemene at 150 μg/mL on gastric cancer cell BGC-823,the effect was mainly characterized by inducing gastric cancer apoptosis at an early stage,the apoptosis rate was(36.9±3.23)%.It showed significant difference compared with the control group(P0.01).Conclusion The Cell-IQ can test and screen the cytotoxicity,cytoactive and cell proliferation.Elemene has obvious effect on inhibition of BGC-823 cell proliferation with the optimum concentration 150 μg/mL.The main mechanism of elemene in inhibiting BGC-823 cell proliferation presumably be the induction of apoptosis rather than cytotoxicity.
