用生物磁粒分离纯化小麦种胚中DNA的研究

摘要 用表面修饰有羧基的生物磁粒对小麦种胚提取液中DNA 的纯化做了系统的研究,得到提取纯化的最佳条件.结果表明,在提取温度50~55℃、提取时间1 h、磁粒用量20 μL条件下,处理15 m g 样品可得到纯度很高的DNA 约43 μg.此法可直接在离心管中操作,具有不离心,不接触氯仿、酚等有机萃取剂等特点. Wheat seeds are treated with an extraction buffer and the supernatants are used to produce pure DNA using carboxyl coated paramagnetic particles. It is proved to be a simple, rapid and efficient method in purification and isolation of DNA. The optimizing condition is incubation temperature:50~55℃, incubation time:1 hour and it can obtain about 43 ug DNA when 15 mg sample is treated. The method for small scale Genomic DNA isolation from plant tissue is simple to perform in micro centrifuge tubes. It eliminates centrifugation and vacuum manifolds, and no toxic phenol/chloroform extractions.
