银翘散体外鸡胚成纤维细胞最大安全质量浓度

摘要 采用体外细胞培养法及四甲基偶氮唑盐MTT比色分析法.研究银翘散及其主要成分在鸡胚成纤维细胞(CEF)体外培养中的最大安全质量浓度.借助光学倒置显微镜,进行肉眼观察细胞的形态,分析药物能否抑制病毒在细胞感染中的增殖现象.MTT法利用活性染料仅对活细胞染色,而受病毒感染破坏的细胞不被染色的原理,作用一段时间后测定光密度吸收OD值,计算出细胞破坏百分率.银翘散的细胞毒性很低,对CEF基本无毒性作用,得出最大安全质量浓度为62.5 mg/mL,而MTT得出的最大安全质量浓度是15.625 mg/mL.高质量浓度(分别大于15.625、15.625、125 mg/mL)时对CEF细胞表现出毒性.在中低质量浓度(分别为7.81 mg/mL、3.906 mg/mL)时对CEF细胞具有较强的促生长和延长存活时间的作用.MTT法除了比形态学观察法更客观、能定量外,还具有更高的敏感性. Using in vitro cell culture method and four methyl azo thiazole salt MTT colorimet- ric analysis to research the maximum security concentration of Yinqiao scattered and its main ingredients in vitro of CEF. Using optical inverted microscope to observe the shape of the cells by naked eye, and analysis whether the drug can restrain proliferation phenomenon of cell infection caused by the virus. The MTT method uses the principle that reactive dyes only dying living cells and the viral infection destruction of cells will not be dying, determine light density absorption OD values after a period of time, then calculate the damage percentage of cell. The cytotoxicity of Yinqiao scattered was very low, almost does not produce toxic effects to CEF. The maximum security concentration was 62.5 mg/mL, but the maximum security concentration was 15. 625 mg/mL by MTT method. At a high concentration ( Respectively than 15. 625, 15. 625, 125 mg/mL) ,it showed that toxicity to CEF ceils, at a low concentration (Respectively was 7.81, 3. 906 mg/mL),it had effect of growth promoting and prolong survival time to CEF cells. MTT method was more objective than morphology observation and could be used, it also had higher sensitivity.
