摘要
目的探讨芳香新塔花黄酮(ZCF)的抗氧化活性,以及对大鼠乳鼠心肌细胞缺氧—复氧损伤的影响。方法采用二苯代苦味酰基自由基(DPPH自由基)清除法测定ZCF的自由基捕获清除能力。体外培养原代SD乳鼠心肌细胞,建立心肌细胞缺氧—复氧损伤模型,用MTT法测定药物对心肌细胞存活率的影响,测定给药后丙二醛(MDA)的变化。结果 ZCF浓度与DPPH自由基的清除率存在量效关系,半数清除浓度IC50为0·017mg/ml。ZCF能明显降低缺氧—复氧模型下培养液中的MDA浓度(P<0·05)。结论 ZCF具有潜在的抗氧化和心肌保护作用。其心肌保护作用机制可能与抑制脂质过氧化,增强心肌细胞抗氧化能力有关。
Objective To study the antioxidant activity and hypoxia/reoxygenation injury protective effects of flavonoids(ZCF) isolated fromZiziphora clinopodioideson neonatal rat cardiomyocytes damaged by hypoxia/reoxygenation stress.Method The removing DPPH experiment was used.The neonatal rat cardiomyocytes were isolated and cultured in vitro to establish hypoxia/reoxygenation injury model.The viability of cardiomyocytes was measured by MTT method.MDA content in medium was also determined.Result There was a good Dose-depandent relationship between ZCF concentration and the clearance rate of DPPH.IC50of ZCF was 0.017mg.ml-1.ZCF can significantly decrease MDAcontent in the model of hypoxia/reoxygenation injury(P0.05).Conclusion ZCF has a potential antioxidant activity and protective effects on neonatal cardiomyocytes damage induced by hypoxia/reoxygenation stress through reduction of lipid peroxidation and enhancement of the activity of antioxidant defense.
