芳香新塔花黄酮抗氧化活性及对大鼠乳鼠心肌细胞缺氧复氧损伤的保护作用 被引量:10

摘要 目的探讨芳香新塔花黄酮(ZCF)的抗氧化活性,以及对大鼠乳鼠心肌细胞缺氧—复氧损伤的影响。方法采用二苯代苦味酰基自由基(DPPH自由基)清除法测定ZCF的自由基捕获清除能力。体外培养原代SD乳鼠心肌细胞,建立心肌细胞缺氧—复氧损伤模型,用MTT法测定药物对心肌细胞存活率的影响,测定给药后丙二醛(MDA)的变化。结果 ZCF浓度与DPPH自由基的清除率存在量效关系,半数清除浓度IC50为0·017mg/ml。ZCF能明显降低缺氧—复氧模型下培养液中的MDA浓度(P<0·05)。结论 ZCF具有潜在的抗氧化和心肌保护作用。其心肌保护作用机制可能与抑制脂质过氧化,增强心肌细胞抗氧化能力有关。 Objective To study the antioxidant activity and hypoxia/reoxygenation injury protective effects of flavonoids(ZCF) isolated fromZiziphora clinopodioideson neonatal rat cardiomyocytes damaged by hypoxia/reoxygenation stress.Method The removing DPPH experiment was used.The neonatal rat cardiomyocytes were isolated and cultured in vitro to establish hypoxia/reoxygenation injury model.The viability of cardiomyocytes was measured by MTT method.MDA content in medium was also determined.Result There was a good Dose-depandent relationship between ZCF concentration and the clearance rate of DPPH.IC50of ZCF was 0.017mg.ml-1.ZCF can significantly decrease MDAcontent in the model of hypoxia/reoxygenation injury(P0.05).Conclusion ZCF has a potential antioxidant activity and protective effects on neonatal cardiomyocytes damage induced by hypoxia/reoxygenation stress through reduction of lipid peroxidation and enhancement of the activity of antioxidant defense.
