秸秆制氢过程中纤维素酶酶活测定方法研究 被引量:6

摘要 秸秆类生物质进行酶解糖化,而后进行光合细菌产氢过程中,确定其纤维素酶酶负荷是秸秆酶法预处理的关键步骤。确定其酶负荷首先要对纤维素酶酶活力进行测定,酶活力的测定有多种方法可以选用。将多种纤维素酶活测定方法进行融合,滤纸酶活力可以代表纤维素酶的总体酶活力,且分光光度计法大大缩短了酶活力测定所需要的时间,且具有较高的精确度,因此最终选择DNS(3-5二硝基水杨酸)分光光度计方法进行试验,所选葡萄糖一次性绘制出线性相关系数达0.999 0的葡萄糖标准曲线,利用一种新型的滤纸酶活计算方法计算并加以验证,通过与其他方法的对比,收到了满意的效果,为确定秸秆类生物质酶解产氢过程中的酶负荷起到了关键作用。 In the process of enzymolysis pretreatment in straw biomass,and the solution in photosynthetic bacteria hydrogen,determining its cellulose enzyme load is the key step.Before enzyme load determining,the enzymatic activity should be determined firstly and a variety of methods can be chosen.In this paper many measuring methods are merged to one,because filter paper enzyme can represent overall cellulase enzyme and spectrophotometer method of enzymatic activity determination greatly can shorten the time required but with higher precision.So DNS spectrophotometer method is finally chosen.The selected glucose test can mapped the 0.9990 linear correlation coefficient of glucose standard curve once.And then using a new type of filter paper enzyme live calculation method to calculate and verify,through comparison with other methods a satisfactory result is received.Overall the method plays a key role in the enzymolysis straw biomass and hydrogen production process of determining the enzyme load.
