摘要 目的通过Morris水迷宫实验,观察成年大鼠单次乳化异氟醚(EI)静脉麻醉后行为学的变化,探讨EI对学习记忆功能的影响。方法成年雄性SD大鼠(n=72),随机分为空白对照组(C组n=12)、30%脂肪乳组(E组n=12)、8%乳化异氟醚组(EI组,内含四个亚组即EI麻醉后2h、1d、7d、14d组,每组n=12)。给药前所有大鼠先进行Mor-ris水迷宫训练6d,记录逃避潜伏期,总路程,平台象限游泳时间,平台象限游泳路程占总游泳路程百分比,游泳速度和经过平台次数。测试结束后,C组不做任何处理、E组给予30%脂肪乳(1.5ml/kg)后2h再次行水迷宫测试);EI组给予8%乳化异氟醚(1.5ml/kg)后按上述分组分别于麻醉苏醒后2h、1d、7d、14d再次行水迷宫测试,观察指标同麻醉前。结果 EI后2h组逃避潜伏期(15.5±15.3)s和总路程(386.8±227.2)cm明显长于C、E组潜伏期(8.8±6.1,9.3±2.8)s和总路程(234..8±118.4,291.6±89.3)cm(P<0.01);EI后2h组平台象限停留时间(35.0±9.8)s与C组平台象限停留时间相比(45.4±12.7)s明显缩短(P<0.01);平台象限游泳路程占总游泳路程百分比、游泳速度和经过平台次数各组组间比较均无显著性差异(P>0.05)。结论单次静脉注射EI在2h内对大鼠学习记忆能力有一定影响,停药1d后大鼠空间学习记忆能力即可恢复到麻醉前水平,提示单次注射EI不会造成持久性的学习记忆功能损害。 Objective To observed behavior changes of the adult rats through Morris water maze test after a single injection of emulsified isoflurane(EI) intravenous anesthesia and investigate the effect of EI on learning and memory in rats.Methods 72 healthy male SD rats weighing 267.7±11.8g were randomly divided into control group(group C n=12),intralipid group(groupE n=12),and emulsified isoflurane group(group EI n=48).The EI group was divided into 4 subgroups(ie,EI anesthesia 2h,1d,7d,14d groups,each group n=12).All rats were trained with Morris water maze test for 6 days consecutively before administered.The escape latency,total distance,swim tine of platform quadrant,the percentage of the distance of platform quadrant and total swim distance,swim speed and number of through platforms were recorded.Results The escape latency(15.5±15.3 s) and the total distance(386.8±227.2 cm) of the EI anesthesia 2h group was significantly longer than the C,E group latency(8.8±6.1,9.3±2.8) s and the total distance(234.8±118.4,291.6±89.3) cm(P0.01).Compared with the C groups,the swim tine of platform quadran in the EI anesthesia 2h group(35.0±9.8) s was significantly shorter(P 0.01),but the percentage of the distance of platform quadrant and total swim distance,swim speed and number of through platforms showed no significant difference between each group(P0.05).Conclusion The single intravenous injection of EI in the 2h has an impact of the learning and memory in adult rats.1d after the withdrawal,the spatial learning and memory of rats can be restored to the level before anesthesia.
