端粒酶活性与甲状腺肿瘤相关性 被引量:2

ANALYSIS OF TELOMERASE ACTIVETY IN THYROID TUMOR

摘要 目的研究端粒酶在各种甲状腺肿瘤(thyroid tumors)中的活性状态。方法用端粒重复序列扩增法(半定量PCR法)在定性及半定量水平上检测88例良恶性甲状腺组织标本及11例甲状腺癌细针穿刺标本的端粒酶活性。结果甲状腺癌组织的端粒酶阳性率(100%)明显高于良性甲状腺病变组织(32%),及病变旁正常甲状腺组织(8.7%);11例甲状腺癌的细针穿刺标本呈阴性表达。半定量结果显示甲状腺癌组织端粒酶活性明显高于良性病变(P<0.01)及正常组织(P<0.05)。结论活性端粒酶是恶性甲状腺肿瘤的一个敏感标志物。半定量PCR分析对甲状腺肿瘤的鉴别诊断具有意义。 Objective To study the status of telomerase activity in different kind of thyroid tumors.Methods Telomerase activity was measured in 11 thyroid carcinomas,54 benign thyroid tumors and 23 normal thyroid tissue samples by means of telomerase half-ration polymerase chain reaction.The assay was also applied to 11 fine needle aspirates(FNAs) of thyroid carcinomas to test its sensitivity.Results All of 11 thyroid carcinomas,17 of 54 benign thyroid tumors,and 2 of 23 adjacent normal thyroid tissue samples expressed telomerase activity,11 FNAs had negative telomerase activity.The half-quantitative analysis showed that the telomerase activity was significantly higher in thyroid carcinomas than those in benign thyroid tumor samples and normal thyroid tissue samples.Conclusion Telomerase activity is a good marker for thyroid carcinomas.The half-ration-PCR analysis used in the differential diagnosis of thyroid neoplasm is of great significance.
