山奈酚对人小细胞肺癌H446的增殖抑制和诱导凋亡作用的影响

摘要 目的探讨山奈酚对人小细胞肺癌H446细胞生长的抑制作用和诱导凋亡作用。方法采用MTT法检测生长的抑制作用;流式细胞仪检测细胞凋亡;Western Blotting技术检测凋亡相关蛋白。结果不同浓度的山奈酚能抑制人小细胞肺癌H446细胞生长,且随浓度增加抑制作用增强(P<0.01)。流式结果显示不同浓度山奈酚组凋亡率较空白对照组均明显增高。Western Blotting结果显示山奈酚处理24 h后,bax蛋白水平升高,而bcl-2蛋白表达降低(P<0.05或<0.01)。结论 山奈酚能抑制人小细胞肺癌H446细胞的增殖并诱导其凋亡。 Objective To investigate the effects of kaempferol on proliferation and apoptosis of human small cell lung cancer H446 cells line in vitro.Methods The MTT assay was used to detect the growth inhibition rates of H446 cells that were cultured by adding Kaempferol into medium in different concentrations.The cell apoptosis was detected by flow cytometry(FCM).The protein expressions of Bax and Bcl-2 were detected by Western Blot.Results The inhibitory effect of kaemperol on H446 cells were significantly increased with the drug concentrations(P0.01),and the effect was in a concentration-dependent manner in vitro.The cell apoptosis rates in different concentrations groups were significantly increased,as compared with those in control group.The detection results tested by Western Blot showed that 24 hours after the H446 cells were treated by kaempferol,the levels of bax were increased,however,the expression of bcl-2 was significantly decreased(P0.05 or 0.01).Conclusion Kaemperol can significantly inhibit the proliferation of H446 cells and can induce cell apoptosis.
