摘要
为了研究在光照和避光条件下姜黄素(CUR)对人胃腺癌BGC-823细胞膜通透性及线粒体膜电位的影响,探讨CUR抑制BGC-823细胞生长的机制,在体外培养的BGC-823细胞加CUR后光照或避光处理24 h,利用激光共聚焦显微镜(LSCM)检测线粒体膜电位变化,利用流式细胞仪(FCM)检测细胞膜通透性改变.结果表明:BGC-823细胞避光组(0、5、20μmol/L)、光照组(0、5μmol/L)的线粒体膜电位分别为(4.71±1.81)、(4.82±1.86)、(0.47±1.42)、(4.26±1.67)、(0.55±1.36),光照5μmol/L CUR组、避光20μmol/L CUR组与相应对照组线粒体膜电位均显著降低;各处理组细胞膜通透性亦发生改变,光照5μmol/L CUR组、避光5μmol/L CUR组膜通透性较对照组均显著升高,且光照5μmol/L CUR组较避光5μmol/L CUR组质膜通透性显著升高.
To investigate the effects of curcumin with or without illumination on mitochondria membrane potential and cell membrane permeability in human gastric cancer cell BGC -823, the BGC -823 cell was treated by curcumin with illumination or in dark. Laser scanning confocal microscope was used to determine mitochondria membrane potential of cell. Flow cytometry was used to investigate cell membrane permeability. The results showed: the changes of mitochondria membrane potential in CUR in dark (0,5,20 μmol/L) and with illumination (0,5 μmol/L) were (4. 71 ±1.81) , (4. 82 ±1.86) , (0. 47 ± 1.42) , (4. 26 ±1.67) , (0. 55 ±1.36) respectively, and mitochondria membrane potential of BGC - 823 cell in 5 μmol/L group treated with illumination was significantly lower than that in the control group and in 5 μmol/L group treated in dark. CUR also induced the significant changes of cell membrane integrity.
