鹿茸及鹿鞭对肾阳虚大鼠不育症的实验研究 被引量:12

Effect of Cornu Cervi Pantotrichum and Penis Cervi on Infertility in Rats with Kidney Yang Deficiency

摘要 【目的】观察鹿茸及鹿鞭对肾阳虚大鼠不育症的影响。【方法】选用清洁级SD大鼠,随机分为6组:正常组,模型组,鹿茸高、低剂量组(剂量分别为0.84、0.21 g.kg-1.d-1),鹿鞭高、低剂量组(剂量分别为3.76、0.94 g.kg-1.d-1);除正常组外,其他组大鼠均采用腺嘌呤(250 mg/kg)灌胃法复制肾阳虚不育症模型。造模成功后各组分别按设计剂量给药,连续5周。末次给药后,称量各组大鼠体质量及附睾、睾丸质量,计算性腺系数;取各组大鼠附睾检测精子总数、活率及顶体酶水平,采用放射免疫法检测各组大鼠血液中促黄体生成素(LH)、睾酮(T)、卵泡刺激素(FSH)水平。【结果】模型组精子总数,活率,顶体酶水平,附睾及睾丸质量、系数,T水平均显著下降,与正常组比较差异有显著性意义(P<0.05或P<0.01);鹿茸及鹿鞭高、低剂量组均可升高精子总数、活率、顶体酶水平、附睾系数,两药高剂量组均可升高睾丸系数及T水平,降低FSH水平,鹿茸高剂量组可升高附睾质量,与模型组比较差异均有显著性意义(P<0.05或P<0.01)。【结论】鹿茸及鹿鞭治疗不育症的作用与其能升高顶体酶水平,改善性激素水平有关。 Objective To observe the effect of Cornu Cervi Pantotrichum(CCP) and Penis Cervi(PC) on infertility in rats with kidney yang deficiency.Methods Clean SD rats were randomized into 6 groups:normal group,model group,high-and low-dosage CCP groups(in the dosages of 0.84 and 0.21 g·kg-1·d-1,respectively),and high-and low-dosage PC groups(in the dosages of 3.76 and 0.94 g·kg-1·d-1,respectively).Except for the normal group,the rats in other groups were given gastric infusion of adenine(250mg/kg) to induce kidney-yang deficiency infertility.After the modeling,the medication groups were given the corresponding drugs for 5 continuous weeks.After the last medication,the body weight,testis index and epididymis index were examined,the sperm count,sperm motility and acrosomal enzyme level in the epididymis were observed,and serum luteinizing hormone(LH),testosterone(T),and follicle-stimulating hormone(FSH) levels were detected in all of the groups.Results In the model group,the sperm count,sperm motility,acrosomal enzyme level,testis weight and index,epididymis weight and index,and T level were decreased(P0.05 or P0.01 compared with those in the normal group).High-and low-dosage CCP and PC increased the sperm count,sperm motility,acrosomal enzyme level,and epididymis weight and index.In high-dosage CCP group and high-dosage-PC groups,testis weight and index as well as T level was increased,FSH level was decreased.High-dosage CCP increased epididymis weight.The differences were significant as compared with those in the model group(P0.05 or P0.01).Conclusion The therapeutic mechanism of CCP and PC for treatment of kidney-yang deficiency infertility is related with the increase of acrosomal enzyme level and with the regulation of sexual hormones.
