摘要
【目的】观察中药抗病毒1号、2号胶囊对单纯疱疹病毒(HSV)-2感染骨髓间充质干细胞(BMSCs)Toll样受体9(TLR9)和干扰素(IFN)表达的影响。【方法】培养BMSCs进行HSV-2病毒接种,分别设立抗病毒1号、2号胶囊组,阿昔洛韦组,病毒组与空白对照组,采用Real-Time RT-PCR方法检测TLR9 mRNA表达,酶联免疫吸附(ELISA)法检测其下游产物IFN-α、IFN-β的表达。【结果】抗病毒1号、2号胶囊组TLR9 mRNA表达升高,IFN-α、IFN-β表达较病毒组显著升高(P﹤0.01),且抗病毒2号组作用优于抗病毒1号组(P﹤0.01)。【结论】抗病毒1号、2号胶囊治疗生殖器疱疹等病毒性疾病作用的可能机制与其能诱导TLR9及其下游产物IFN-α、IFN-β表达升高有关。
Objective To study the effect of Antiviral Capsule No.1 and No.2 on Toll-like receptor 9(TLR9) expression and interferons(IFN) in mesenchymal stem cells of bone marrow(BMSCs) infected with herpes simplex virus 2(HSV-2).Methods The cultured BMSCs were inoculated with HSV-2,and then were divided into Antiviral Capsule No.1 group,Antiviral Capsule No.2 group,acyclovir group,HSV-2 group and blank control group.Real-time RT-PCR was used to detect the expression of TLR9 mRNA,and enzyme-linked immunosorbent assay(ELISA) was used to detect the expression of interferon alpha(IFN-α)and IFN-β.Results The expression of TLR9 mRNA,IFN-α and IFN-β were increased in Antiviral Capsule No.1 group and Antiviral Capsule No.2 group(P0.01 compared with that in HSV-2 group),and the increase of the indexes was obvious in Antiviral Capsule No.2 group(P0.01 compared with that in Antiviral Capsule No.1 group).Conclusion The therapeutic mechanism of Antiviral Capsule No.1 and No.2 for viral diseases such as genital herpes is probably related with the induction of the expression increase of TLR9 and its downstream products of IFN-α and IFN-β.
