阳春砂愈伤组织诱导的多因子正交试验研究 被引量:3

摘要 【目的】优化阳春砂愈伤组织诱导方法,以提高愈伤组织诱导率。【方法】采用正交设计法,观察以未成熟及成熟种子为外植体,以及培养基中添加的不同植物生长物质对阳春砂愈伤组织诱导的影响。【结果】最佳培养基为MS+二氯苯氧乙酸(2,4-D)(0.5 mg/L)+6-苄基腺嘌呤(6-BA)(2 mg/L)+AgNO3(6 mg/L)+酸水解酪蛋白(1 mg/L),植物生长物质中2,4-D对阳春砂愈伤组织的形成影响最关键,未成熟的阳春砂种子是最佳外植体,愈伤组织诱导率最高达到65.41%。【结论】植物生长物质中2,4-D对阳春砂愈伤组织的形成不可或缺,成熟程度低的阳春砂种子更适宜作为愈伤组织诱导的外植体。 Objective To optimize the method for callus induction in Amomum villosum Lour.(AVL).Methods Effects of different explants and different kinds of medium on callus induction in AVL were investigated by orthogonal design.Results The optimal medium was MS with 0.5 mg·L-1 of 2,4-dichtorophenoxyacetic acid(2,4-D),2 mg·L-1 of 6-Benzylaminopurine(6-BA),6 mg·L-1 of AgNO3,and 1 mg·L-1 of acid hydrolysis casein added.Plant growth substance of 2,4-D was the key for callus induction in AVL.The optimal explant was the immature AVL seed.With the immature AVL seed as the explant and with the optimal medium as the culture medium,the highest rate of callus induction in AVL was up to 65.41%.Conclusion 2,4-D is critical for callus induction in AVL,and immature AVL seeds are suitable for callus induction in AVL.
