摘要
用放射免疫测定法(RIA)和吖啶橙染色法分别检测31例SLE患者和22名正常对照血清脱氢表雄酮(DHEA)水平和其在体外对淋巴细胞凋亡的影响。发现SLE患者血清DHEA水平较正常对照明显降低 ,淋巴细胞凋亡率明显升高 ;生理浓度的DHEA在体外能减少SLE患者淋巴细胞的凋亡。结果提示SLE患者体内淋巴细胞凋亡率高于正常 ,且DHEA水平的低下可能是导致SLE患者淋巴细胞凋亡率升高的原因之一。
The serum levels of dehydroepiandrosterone (DHEA) and the rates of in vitro spontaneous apoptosis of peripheral blood lymphocytes incubated with or without DHEA in 31 SLE patients and 22 normal controls were assayed by radioimmunoassay (RIA) and acridine orange(AO) staining method respectively. The results showed that the DHEA levels in SLE patients were significantly lower than those in normal controls, and that fresh lymphocytes from SLE patients demonstrated a significant increase in the number of apoptotic cells compared with those of normal subjects. Furthermore, physiological concentration of DHEA was able to reduce in vitro apoptosis of lymphocytes derived from SLE patients. The above findings suggest that there may be a higher rate of in vivo lymphocyte apoptosis in SLE patients. Low serum level of DHEA in SLE may be one of the causes for the increased rate of apoptosis in vivo.
