类胰岛素生长因子1对转化生长因子β_1诱导前列腺增生间质细胞类胰岛素生长因子结合蛋白3生成的抑制作用 被引量:1

The inhibitory effects of insulin like growth factor 1 on the induction of IGFBP 3 in prostate stromal cell by TGFβ_1

摘要 目的 探讨类胰岛素生长因子(IGF1) 和转化生长因子(TGFβ1) 在调节前列腺间质细胞IGF结合蛋白———类胰岛素生长因子结合蛋白3(IGFBP3) 中的作用。方法 前列腺增生组织原代培养分离间质细胞。分别用TGFβ1(5 μg/L)、IGF1(500 μg/L) 及TGFβ1+ IGF1 处理间质细胞。Western blot检测间质细胞内的IGFBP3 水平。结果 间质细胞用TGFβ1 处理2 小时后,IGFBP3 比对照组增加132 % ,4 小时后比对照组高出251% ,8 小时后接近原来水平。IGF1 可以明显逆转TGFβ1 诱导增加的IGFBP3,而单纯IGF1 则对间质细胞的IGFBP3 水平没有明显的影响。结论 IGF1 有抑制TGFβ1 诱导前列腺间质细胞IGFBP3 表达的作用。 Objective\ To investigate the role of insulin like growth factor 1 (IGF1) and TGFβ1 in the regulation of IGFBP 3 in prostate stromal cells. Methods\ Primary culture stromal cells derived from benign prostate hyperplasia patients were treated with TGFβ1 (5 ng/ml), IGF1 (500 ng/ml) and TGFβ1 plus IGF1 respectively. Western blot was used to detect the expression of IGRBP 3 in the stromal cells. Results\ Compared to the control cells IGFBP 3 was increased by 132 % at 2 h and 251 % at 4 h following treatment with TGFβ1. Increasing IGRBP 3 level induced by TGFβ1 was reversed by IGF1 but IGF1 alone had no significant effect on the IGFBP 3 level in prostate stromal cells. Conclusion\ IGF1 can significantly inhibit the induction of IGFBP 3 in prostate stroma cells by TGFβ1.\;
