摘要
目的:考察知母总多糖(TPA)的抗炎作用,并初步探讨其作用机制。方法:应用二甲苯致小鼠耳廓肿胀、醋酸致小鼠腹腔毛细血管通透性增高、角叉菜胶致大鼠足趾肿胀及大鼠棉球性肉芽肿增生动物模型进行整体药效试验。结果:TPA能显著抑制上述各种动物模型的炎症反应,但不能显著抑制摘除双侧肾上腺大鼠角叉菜胶性足趾肿胀;TPA能显著提高大鼠血浆中皮质酮浓度,而使血浆中ACTH 浓度显著降低;TPA 还能显著降低炎症组织中PGE含量。结论:TPA对急慢性炎症均有明显的抑制作用,促进肾上腺分泌较高水平的糖皮质激素及抑制炎症组织PGE的合成或释放是其发挥抗炎作用的重要途径。
Objective: To investigate the anti inflammatory activity of total polysaccharides of Anemarrhena asphodeloides Bunge (TPA) and discuss the mechanism roughly. Methods: We observed activities shown in various models of experimental inflammation including: (1)mouse ear inflammation caused by xylen;(2) increased vascular permeability induced by acetic acid in mice;(3) paw edema caused by carrageenin in rats; (4) granuloma caused by implanted cotton ball in rats. Results: Anti inflammatory activities of TPA were found in all those models above, but anti inflammatory activity wasn′t found in rats whose adrenal glands were extirpated. Compared with control group, corticosterone level in plasma of TPA group increased obviously, while ACTH level decreased, PGE level in inflammatory tissue also lowered. Conclusion: TPA could be used to cure not only acute inflammation but also chronic inflammation in animal, and promoting the adrenal gland to secrete glucocorticoid and inhibiting the synthesize and release of PGE in inflammatory tissue might be two important routes of TPA.
