摘要
目的观察养心透毒方对柯萨奇病毒(CVB3)感染小鼠造成的CVB3病毒性心肌炎(VMC)的保护作用。方法将HeLa细胞培养的CVB3制成悬液,测定半数组织细胞感染量(TCID50),小鼠腹腔接种CVB3培养液建立VMC模型。小鼠随机分为正常组,模型组,利巴韦林+辅酶Q10(CoQ10)组,养心透毒方低、高剂量组,灌胃给药4周。应用酶联免疫法检测各组小鼠IgG和IgM阈值,测定血清肌钙蛋白T(cTnT)、白细胞介素-2(IL-2)、肿瘤坏死因子-α(TNF-α)水平,组织匀浆测定心肌IL-2和TNF-α含量,HE染色观察小鼠的心肌组织病理形态学改变。结果养心透毒方低、高剂量组IgM、IgG阳性的动物数均少于模型组。模型组心肌组织及血清IL-2、TNF-α含量与正常组比较均有显著性差异(P<0.05),心肌组织有明显病理改变。养心透毒方可使小鼠心肌组织坏死程度减轻,心肌炎症浸润减少,降低组织和血清的IL-2和TNF-α含量(P<0.05)。结论养心透毒方能减少小鼠CVB3病毒的感染率,降低组织中炎症因子的水平,对小鼠早期VMC具有防治作用。
Objective To observe the protective effect of Yangxin Toudu Formula(YTF), which is composed of Chinese herbs with the actions of supplementing qi and nourishing yin, activating blood circulation and detoxicating, on mice viral myocarditis (VMC) induced by coxsackie virus B3 (CVB3). Methods We cultivated CVB3 with HeLa cells and made suspensions, and then measured 50 % tissue culture infective dose (TCID50) of the suspensions. Seventy five ICR mice were randomly divided into 5 groups, 15 in each group. The 5 groups were normal group, model group, ribavirin+ Coenzymum Q10 (CoQlO) group, low-dose YTF group and high-dose YTF group. Model group and medication administration groups were injected with CVB3 to induce VMC. After intragastric administration for 4 weeks, serum levels of IgG, IgM, troponin T(cTnT), interleukin 2 (IL-2), and tumor necrosis factor alpha (TNF-α) , and myocardial contents of IL-2 and TNF-α were measured with enzyme-linked immunosorbent assay (ELISA). Meanwhile, the pathomorphological changes of the myocardium were observed by HE staining method. Resuits After 4 weeks, the number of mice with IgM and IgG positive in high- and low-YTF groups was lower than that in the model group, serum and myocardial levels of IL-2 and TNF-α were also significantly different as corn pared with those in the normal group(P 〈 0.05). Moreover, the myocardial pathological changes were obvious in the model group. Compared with the model group, the myocardial necrosis and inflammatory infiltration were significantly improved, and the serum and myocardial levels of IL-2 and TNF-α were also decreased in YTF groups (P 〈 0.05). Conclusion YTF can reduce the infective rate of CVB3, decrease the myocardial levels of inflammatory factors, indicating that YTF has the protective effect on CVB3-induced VMC in mice.
