黄芪多糖对哮喘小鼠气道高反应性及气道重塑的影响 被引量:18

Effect of Astragalus Polysaccharides on Airway Hyperresponsiveness and Airway Remodeling in a Murine Model of Chronic Asthma

摘要 目的观察黄芪多糖对哮喘小鼠气道高反应性及气道重塑的影响。方法通过腹腔注射和雾化吸入卵蛋白(OVA)制备哮喘缓解期小鼠模型。使用黄芪多糖治疗2月后,末次给药后24 h观察小鼠气道反应性、肺泡灌洗液(BALF)中炎性细胞总数和细胞分类以及肺脏病理学变化。结果黄芪多糖可以降低模型小鼠的气道高反应性,降低BALF中炎性细胞总数及嗜酸性粒细胞和中性粒细胞比例,改善支气管周围炎性细胞浸润、减轻气道壁胶原沉积和黏液分泌,减轻气道重塑。结论黄芪多糖降低气道高反应性、减轻支气管周围嗜酸性粒细胞浸润以及改善气道重塑可能是其治疗哮喘的主要作用机制。 Objective To observe the effect of astragalus polysaccharides (APS) on hyperresponsiveness and airway remodeling in mice with chronic asthma. Methods BALB/c mice were sensitized with intraperitoneal injection of ovalbumin (OVA) to induce mice models of asthma in paracmasia. The intragastrical treatment of APS lasted two months, and 24 hours after the last administration, the airway hyper-responsiveness, inflammatory cell count and cell classification in bronchoalveolar lavage fluid(BALF) , and lung histological examination were carried out. Re- suits After APS treatment, the airway hyper-responsiveness was markedly relieved, inflammatory cell count and the ratio of eosinophil and neutrophils in BALF were reduced, inflammatory cell infiltration was relieved, and collagen deposition and mucus production were reduced. Conclusion The therapeutic mechanism of APS for asthma might be related with the decrease of airway hyperresponsiveness, the relief of eosinophiles infiltration and the improvement of airway remodeling.
