摘要
目的观察拟apoE在SAH后对Ctyc、Caspase-3表达的影响,探讨apoE的脑保护可能机制,以期为临床治疗提供理论依据。方法雄性LCR小鼠随机分为对照组、SAH组、apoE治疗组。观测7个时相点:10 min、30 min1、h、3h2、4 h4、8 h7、2 h。采用免疫组织化学法观察Cytc、Caspase-3的表达情况及TUNEL免疫组织化学检测细胞凋亡数。结果 SAH组相比,apoE治疗组Ctyc、Caspase-3阳性细胞免疫阳性反应明显降低,apoE治疗组细胞凋亡数有所减少。结论拟apoE可能是通过降低Cytc的释放减少Caspase-3途径的凋亡而发挥其脑保护作用。
Objective To investingate the effect of apoE on the expressions of Ctyc and Caspase-3 in the rats following SAH.Discuss the possible cerebral-protective mechanism of apoE.Hoping to offer theoretic evidences for clinical therapy and new method of therapy.Methods Male LCR mice were randomly divided into 3 groups:control group,SAH group,and SAH + apoE1410 group.Each group was divided into 7 subgroups according to time phase after brain injury,which was 10min,1h,3h,6h,24h,48h and 72 h.The expression of Ctyc and Caspase-3 was measured by immunohistochemistry and study the number of apoptosis through TUNEL immunohistochemistry.Results Compared with SAH groups,the expressions of Ctyc and Caspase-3 reduced obviously in apoE treatment group.The number of the tunel positive cells obviousely decreased treated by the apoE compared with SAH groups.Conclusion ApoE maybe exert neuroprotective effect through inhibiting cytc releasing lead to decreasing apoptosis of caspase-3 pathway.
