酸敏感离子通道1a在糖尿病鼠局灶脑缺血中的变化及意义

摘要 目的通过检测糖尿病鼠局灶脑缺血不同时间缺血周围皮层酸敏感离子通道1a(Acid-sensing ion channel1a,ASIC1a)表达变化,探讨其在糖尿病局灶脑缺血中的意义。方法 Wistar雄性大鼠72只,应用高脂饮食联合链脲佐菌素STZ制备糖尿病大鼠模型,线栓法制备单纯局灶脑缺血模型及糖尿病大鼠局灶脑缺血模型,根据缺血不同时间分为正常假手术组、单纯脑缺血1 h,3 h,6 h,24 h;糖尿病脑缺血1 h,3 h,6 h,24 h共计9组,每组8只。应用TTC染色及神经功能评分比较各组梗死面积及损伤程度;Western Blot法检测各组缺血周围皮层的蛋白表达。结果对糖尿病局灶脑缺血24 h及单纯缺血24 h大鼠进行神经评价,糖尿病组大鼠术后清醒较晚,肢体偏瘫程度重于单纯缺血组(2.60±0.591/1.80±0.837),两组相比差异显著(P<0.05);与正常假手术组相比,随着缺血时间的延长,单纯局灶脑缺血及糖尿病局灶脑缺血各组ASIC1a的表达逐渐增多(P<0.05),其中糖尿病局灶脑缺血各组ASIC1a的蛋白表达高于单纯局灶脑缺血各组(P<0.05)。结论糖尿病局灶脑缺血加重的原因可能由于皮层ASIC1a过度开放加重细胞内钙超载所致。 Objective After measuring the change of Acid-sensing ion channel 1a during focal brain ischemic in rats(MCAO) and diabetes mellitus rats(DM+MCAO) with different time,investigate its significance in focal brain ischemic in diabetes mellitus rats.Methods 72 Wistar male rats,combine high fat diet with STZ for establishing DM model,,use thread embolism to prepare MCAO and DM+MCAO model.Experiment divided:normal sham operation,MCAO1 h、MCAO3 h、MCAO6 h、MCAO24 h、DM+MCAO1 h、DM+MCAO3 h、DM+MCAO6 h、DM+MCAO24 h.nine groups and 8 rats per group;,TTC dyeing to identificate ischemic brain tissue,nerve function score judge the degree of palsy with MCAO24 h and DM+MCAO24 h and compare them.,measure the change of Acid-sensing ion channel 1a around ischemic pyramidal cell of cortex in each group with Western Blot.Results DM+MCAO 24 h rats awake slowly,degree of semiplegia is more serious than MCAO 24 h,score of function nerve of DM+MCAO is higher than MCAO(2.60±0.591/1.80±0.837),significant difference(P0.05).:with the longer of ischemic time,the more of ASIC1a during MCAO and DM+MCAO among them(P0.05),each group of DM+MCAO is higher than each group of MCAO respectively(P*0.05).Conclusion the reason of aggratation during DM+MCAO is probably associated with ASIC1a open and Ca2+ overloaded.
