将构建的２ 种重组表达质粒ｐＳＶ２ｕＰＡ和ｐｃＤＮＡ３ｕｐＡ分别直接注入小鼠骨骼肌，观察了ＳｃｕＰＡｃＤＮＡ 在小鼠体内的表达情况－ 体外凝血试验显示注射质粒ＤＮＡ 后，第５ ｄ 凝血时间有所延长，第１５ ｄ 凝血时间延长幅度最大，较对照组平均延长６ ｍｉｎ 左右；第２０ ｄ 体内诱导产生ＤＩＣ，体外凝血时间试验组较对照组延长一倍，抗凝效果明显，２ 种表达质粒在体内的表达水平无明显差异－

Two expression plasmids,pSV 2 uPA and pcDNA 3 uPA,containing the single chain Urokinase type plasmoneign activator (Scu PA) cDNA gene were constructed.The Scu PA expressed in mouse muscle was observed by direct injection of pSV 2 uPA and pcDNA 3 uPA into the muscle.The result showed that the prolongation of blood clotting time could be discerned at 5th day after the injection of the recombinant plasmids,and the peak occurred on the 15th day.On an average,the blood coagulation time prolonged for 6 min compared with the CK.Disseminated intravascular Coagulation (DIC) was induced on the 20th day.Compered with normal mouse,blood clotting time doubled,and there was no obvious difference between the expression levels of the two plasmids.
