摘要
目的:研究板栗毛壳水提液(AEICM)对大鼠全脑缺血再灌注损伤的保护作用。方法:采用板栗毛壳水提液(AE-ICM)对大鼠连续灌胃7d后,利用4-血管阻断(4-VO)法建立全脑缺血再灌注动物模型,采用干湿质量法观察板栗毛壳水提液(AEICM)对脑水肿的影响,利用分光光度法检测大鼠脑、肝、肾、心等组织中丙二醛(MDA)水平、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-PX)活性。结果:AEICM能显著减轻脑水肿(P<0.05),降低MDA(P<0.01)的水平,增加各器官组织中SOD(P<0.01或P<0.05)和(GSH-PX)(P<0.05)活性。结论:AEICM对脑缺血再灌注损伤有保护作用,其机制可能与抗脂质过氧化有关。
OBJECTIVE To study the protective effects of aqueous extract of Involucrum Castaneae Mollissimae (AEICM) on the whole cerebral ischemia-reperfusion injury in rats. METHODS Thirty mice were randomly divided into the normal group, model group and AEICM group. AEICM was given to the AEICM group orally. After 7 days, the whole cerebral ische- mia reperfusion injury was established by four-vessel occlusion (4-VO). Dry-wet weight method was carried out to study the protective effect of AEICM on brain edema in rats. The activities of SOD and GSH-PX and the level of MDA in the brain, heart, liver, kidney tissues were determined by the ultraviolet spectrophotometer. RESULTS Compared with the model group, the activities of SOD and GSH-PX (P〈0. 01, P〈0. 05) were enhanced remarkably by AEICM in all tissues, but the water content of brain tissue and the level of MDA were decreased remarkably (P〈0. 05). CONCLUSION AEICM had protective effects on the whole cerebral ischemia reperfusion injury in rats, and the mechanism may relate to its inhibitory effect on the lipid peroxidation.
