大花君子兰总生物碱抗肿瘤作用的研究 被引量:2

摘要 采用MTT法观察大花君子兰总生物碱(简写为TACMR)对体外培养的人肺癌细胞NCI-H446、人胃癌细胞SGC-7901、人乳腺癌细胞MCF-7和人肝癌细胞SMMC-7721 4种细胞株的增殖抑制作用;用小鼠移植性肿瘤法检测TACMR对小鼠H22肝癌的生长抑制作用,并通过脾指数和胸腺指数等指标综合考察TACMR对肿瘤小鼠免疫机能的影响。体外MTT结果显示TACMR对NCI-H446、SGC-7901、MCF-7和SMMC-7721均具有明显的细胞增殖抑制作用,均呈现良好的量效关系,并且TACMR作用于4种不同人肿瘤细胞48 h的IC50值分别为38.55,12.08,8.85,7.04μg/mL;体内试验结果表明TACMR对H22肝癌生长具有一定的抑制作用。TACMR剂量在34 mg/kg时,抑瘤率为35.2%,脾指数与阳性对照组相比显著增加(P<0.01)。TACMR剂量在136 mg/kg时,抑瘤率仅为3.7%,与阴性对照组比较,胸腺指数和脾指数明显减少(P<0.01),与阳性对照组比较,胸腺指数明显减少(P<0.01)。上述体内外试验结果表明:TACMR具有抗肿瘤作用,同时对免疫功能有一定的影响。 In vitro, the inhibitory effects of total alkaloids from Clivia miniata Regel (TACMR) on prolif- eration of NCI - H446, SGC - 7901, MCF - 7, and SMMC - 7721 were measured by MTT colorimetric assay. The inhibitory effects of TACMR on H22hepatic cancer was observed in mice with subcutaneous inoculation on the tumor cells and its effect on immunological function of mice bearing tumor by observing the influence to spleen index, thymus index and so on. In vitro, the results of MTT showed that TACMR inhibited proliferation of NCI - H446, SGC - 7901, MCF - 7, SMMC - 7721 and a good dose-effect relationship and the IC50 values on them were 38.55, 12.08, 8.85 and 7.04 ttg/mL. In vivo, TACMR has significant inhibitory effect on H22 tumor ceils. The inhibition ratio was 35.2% with the dosage of 34 mg/kg, compared with the positive control group, spleen index increased significantly (P 〈 0.01 ). The inhibition ratio was 3.7% with the dosage of 136 mg/kg, compared with negative control group, thy- mus index and spleen index decreased significantly ( P 〈 0.01 ), compared with the positive control group, thymus index decreased significantly( P 〈 0.01). In tumor inhibitory test, it was shown that TACMR has strong inhibition on tumor cells and certain impact on immunological function.
