福建省50株沙门菌PFGE指纹图谱研究 被引量:3

Study on PFGE fingerprint of 50 Salmonella strains in Fujian,China

摘要 目的分析福建省鼠伤寒及肠炎沙门菌脉冲场凝胶电泳(PFGE)指纹图谱,研究其分型特征。方法收集2009年50株沙门菌临床分离株,其中30株鼠伤寒沙门菌分离自婴幼儿腹泻患者,20株肠炎沙门菌分离自感染性腹泻患者。选择XbaⅠ酶对沙门菌染色体DNA进行酶切,H9812作为脉冲场凝胶分子量标准。采用PFGE技术分析电泳酶切指纹图谱,运用BioNumerics软件进行聚类分析。结果 30株婴幼儿腹泻鼠伤寒沙门菌PFGE图谱按照100%的相似度可分为19个PFGE型,P1(Typhi)型有8株,P12(Typhi)型3株,P1(Typhi)为优势型别;20株肠炎沙门菌PFGE图谱按照100%的相似度可分为10个PFGE型,其中P1(Enteri)型6株,P4(Enteri)型有4株,P6(Enteri)型3株,P1(Enteri)型为优势型别。结论 PFGE分型方法分辨率高,重复性好,可广泛运用于沙门菌疫情溯源和早期防控。 Objective To analyze PFGE fingerprint of S.Typhimurium and S.Enteritidis in Fujian and study the PFGE typing characteristics.Methods Totally 30 strains of S.Typhimurium from infantile diarrhea patients and 20 strains of S.Enteritidis from infectious diarrhea patients were collected in 2009.DNA was digested by XbaⅠwith H9812 as the molecular weight marker,PFGE fingerprints were analyzed and clustering analysis was carried out by BioNumerics software.Results Taking 100% similarity as criterion,30 strains of S.Typhimurium were classified as 19 PFGE types,in which 8 strains of P1(Typhi) and 3 strains of P12(Typhi).P1(Typhi) was the dominant type.20 strains of S.Enteritidis were classified as 10 PFGE types,in which 6 strains of P1(Enteri),4 strains of P4(Enteri) and 3 strains of P6(Enteri).P1(Enteri) was the dominant type.Conclusion The method of PFGE typing shows high resolution and better reproducibility.It can be generally applied to the source investigation and the disease control and prevention.
