摘要
利用热处理与茎尖培养相结合的方法,脱除姜体内危害性病毒TMV 和CMV,并通过试验研究明确了姜组培适宜的初代接种灭菌剂是HgCl2(0 .1 % ) ;初代培养基为MS+ KT2 .0(mg/L,下同) + NAA1 .0 + 糖3 % + 琼脂0 .5 % ;组培快繁、生根培养的外源激素种类及浓度分别为KT2.0+ NAA0 .5 ,KT1.0+ NAA1 .5 ;培养基的pH值为6 .5 ,培养温度(26±2) ℃,光强4000lx;适宜的组培快繁方式为试管微型姜快繁法。
TMV and CMV in ginger can be taken off through heat treatment and stem apex culture.The bactericide is HgCl 2(0.1%).The first culture medium is MS+KT 2.0 mg/L+NAA 1.0 mg/L+cane sugar 3%+agar 0.5%.The rapid propagation and root promoting hormones are KT 2.0 mg/L+NAA 0.1 mg/L and KT 1.0 mg/L+NAA 1.5 mg/L respectively.The pH of culture medium is 6.5,cultur temperature is (26±2)℃,and intensity of illumination is 4000 lx.The optimum tissue culture method is tube miniature ginger propagation.
