CD、PTEN在胶质瘤中的表达与侵袭性的关系 被引量:3

The relationship of PTEN and CD expressions with invasion in human glioma

摘要 目的探讨组织蛋白酶D(Cathepsin D,CD)和第10号染色体磷酸酶和张力蛋白同源丢失性基因(phosphatase and tension homolog deleted on chromosome10,PTEN)表达与人胶质瘤恶性程度和侵袭性之间的关系。方法采用免疫组织化学法和Western blot法检测46例胶质瘤标本和10例正常脑组织标本中CD及PTEN的表达情况。结果 CD阳性染色主要定位于细胞质内,呈棕黄色细颗粒状,在高分化组(Ⅰ-Ⅱ级)中呈散在的弱阳性表达,表达率为50%(14/28);而在低分化组(Ⅲ-Ⅳ级),表达明显增强,表达率为83.3%(15/18),与高分化组有显著差异(P<0.05)。PTEN阳性染色主要定位于细胞质内,46例胶质瘤中PTEN阳性表达18例(39.1%),高分化组(Ⅰ-Ⅱ级)中15/28例(53.6%)明显高于低分化组(Ⅲ-Ⅳ级)3/18例(16.7%,P<0.05)。结论 CD和PTEN的表达对分析、判断胶质瘤分级及恶性转化具有一定意义。 Objective To study cathepsin-D(CD) and phosphatase and tension homolog deleted on chromosome 10(PTEN) expressions in human glioma,and to investigate the relationship between expression and invasion in human glioma.Methods The expressions of CD and PTEN were observed by immunohistochemical method and Western blot in 46 formalin-fixed paraffin embedded glioma sections and in 10 normal brain tissue.Results The CD positive staining was located in plasma,the positive rate (50%,14/28) in high differentiation group(grade Ⅰ and Ⅱ) was lower than that in low differentiation group(grade Ⅲ and Ⅳ,83.3%,15/18,P〈0.05).The PTEN positive staining was found in plasma,the positive rate was 39.1%(18/46) with higher in high differentiation group than in low differentiation group (P〈0.05).Conclusion Expressions of CD and PTEN are of significance in evaluating the pathological grades and biological behavior of human glioma.
