藤茶对非酒精性脂肪性肝病的治疗作用研究

Therapeutic Effects of Ampelopsis Grossedentata on Nonalcoholic Fatty Liver Disease in Mice

目的研究藤茶对非酒精性脂肪性肝病(NAFLD)小鼠的治疗作用。方法将48只雄性昆明小鼠随机分为4组,各12只。正常组普通饲料喂养;模型组以复方高脂饮食建立NAFLD小鼠模型;阳性对照组在模型组喂养的基础上从第14天开始给予易善复150 mg/kg,治疗42 d;藤茶治疗组在模型组喂养的基础上从第14天开始给予藤茶300 mg/kg,治疗42 d。实验第60天处死小鼠,比较4组小鼠的肝脏形态学及肝指数,血清总胆固醇(TC)、三酰甘油(TG)、丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)水平,肝脏TC、TG含量,肝脂肪变性程度。结果藤茶治疗42 d后,NAFLD小鼠的肝指数,血清TC、TG、ALT、AST水平,以及肝脏TC、TG含量均较模型组显著降低(P<0.05),且与阳性对照组比较差异无统计学意义(P>0.05)。光镜观察结果表明,藤茶治疗组小鼠肝脂肪变性程度较模型组显著减轻(P<0.05),与阳性对照组比较差异无统计学意义(P>0.05)。结论藤茶具有降脂及治疗NAFLD的作用。

Objective To investigate the therapeutic effects of ampelopsis grossedentataon nonalcoholic fatty liver and the possible mechanisms in mice.Methods A total of 48 male mice were randomly divided into four groups：control group,model group,positive control group,and ampelopsis grossedentata group.The model group received high fat diet.The positive control group received polyene phosphatidylcholine（150 mg/kg） for 42 days from the 14th day.The ampelopsis grossedentata group were treated with ampelopsis grossedentata（300 mg/kg） for 42 days from the 14th day.Morphological changes,liver indices,serum total cholesterol（TC）,serum triglyceride（TG）,alanine transaminase（ALT）,aspartate transaminase（AST）,liver TC,liver TG,and degrees of liver fatty degeneration were compared among the 4 groups.Results After treatment with ampelopsis grossedentata,serum levels of total cholesterol（TC）,triglyceride（TG）,alanine transaminase（ALT）,aspartate transaminase（AST）,liver TC,and liver TG were significantly decreased as compared with the nonalcoholic fatty liver model group（P0.05） and showed no significant differences when compared to the positive control group（P0.05）.Histological evaluation of mice liver showed dramatically decreased lipid accumulation in the ampelopsis grossedentata group.Conclusion Ampelopsis grossedentata exhibits therapeutic effect on nonalcoholic fatty liver in mice.
