用抗雄性特异性单克隆抗体鉴别小鼠胚胎性别

摘要 用实验室筛选的雄性特异性单克隆抗体鉴别小鼠胚胎性别时,阳性符合率为67.31% ,阴性符合率为82.22% 。将胚胎在单抗中孵育的时间从105~120 min 减至45 m in,相应地减少了难以分辨雌雄的荧光中间型胚胎数量。为了统一对胚胎中荧光斑的描述,根据荧光组成的基本形状和强度将胚胎荧光型分为8种,按荧光型的不同将胚胎判为阴性和阳性,并逐一用PCR法检测胚胎的真实性别,以检验免疫学鉴别胚胎的符合率,从而建立了更符合客观实际的免疫学鉴别胚胎的标准,提高了小鼠胚胎性别鉴别的效率。 We isolated 9 cell lines male specific antigen monoclonal antibodies(McAbs) to sex mouse embryos,67.13% fluorescence embryos are male and 82.22% nonfluorescence are female examined by PCR.The period of incubating in McAbs is cut down from 105~120 min to 45 min, the number of inter fluorescence embryos that difficult to sex is decreased relevant. To standardize description the fluorescence spots inside the embryos, we have classified the type of embryos fluorescence into 8 basic types according to its shape and its intensity, then according to its shape and intensity divided them into fluorescence and non fluorescence, at last examination all embryos by PCR one by one.Comparing the embryos fluorescence with its truthful sex, we have built a objective standard to sex embryos by immune method, improving the sexing efficiency.
