Matrilin-3基因表达在骨肉瘤转移中的作用

Role of Matrilin-3 gene expression in osteosarcoma metastasis

摘要 目的寻找骨肉瘤转移相关基因,初步分析Matrilin-3基因表达与骨肉瘤转移的关系。方法通过RT-PCR、Westen-blot及免疫组化检测该基因在不同骨肉瘤细胞系、正常成骨细胞、骨肉瘤及正常骨组织临床标本中的表达,并分析其与骨肉瘤患者预后的关系。结果 Matrilin-3在高转移MG36-A1细胞系中表达明显下降。RT-PCR、Westen-blot检测发现Matrilin-3在正常成骨细胞中高表达,在SAOS2、143B中表达缺失。免疫组化检测显示在25例骨肉瘤临床标本中,24%(6/25例)的标本Matrilin-3表达为阳性,其余为阴性;在正常骨组织中Matrilin-3表达强阳性。Matrilin-3表达与骨肉瘤患者预后呈正相关(P=0.023)。结论 Matrilin-3的表达随着骨肉瘤细胞系转移能力的增加而出现缺失,Matrilin-3可能成为阻遏骨肉瘤转移的新靶点。 Objective:To preliminarily analyze the relationship of Matrilin-3 gene expression and metastasis of osteosarcoma through the whole of osteosarcoma metastasis related genes.Methods By RT-PCR,Westen-blot and immunohistochemistry to detect the expression of the gene in the different osteosarcoma cell lines,normal bone cells,osteosarcoma and normal bone tissue clinical specimens,and to analyze the relationship of the prognosis of patients with osteosarcoma.Results Matrilin-3 expression decreased significantly in the cell line of MG36-A1.RT-PCR,Westen-blot test confirmed Matrilin-3 high expression in normal osteoblasts and the loss of expressionin inSAOS2,143B.Immunohistochemistry displayed in 25 cases of osteosarcoma clinical samples,24%(6/25 cases) specimens Matrilin-3 were expressed as positive,the rest were negative;in normal bone tissue Matrilin-3 expression strongly positive.Matrilin-3 expression and prognosis in patients with osteosarcoma were positively correlated(P=0.023).Conclusion Matrilin-3 expression of deficiencies in osteosarcoma cell lines arising from the increased transfer capacity.Matrilin-3 may be to deter a new target for osteosarcoma metastasis.
