摘要
目的:探讨当归注射液治疗对帕金森病的可能作用及作用机制。方法:采用6-羟基多巴胺(6-OHDA)处理PC12细胞制作帕金森病体外模型。6-OHDA处理前2h,将当归注射液加入PC12细胞培养液中,用四甲基偶氮唑蓝(MTT)法检测细胞存活率,采用流式细胞仪检测凋亡率,并用免疫细胞化学SABC法和图像分析检测酪氨酸羟化酶(TH)及SIRT1表达。结果:高浓度当归注射液能明显提高PC12细胞的存活率(P<0.01),增加TH阳性细胞数(P<0.01),降低细胞凋亡率(P<0.01),增加SIRT1表达(P<0.01)。结论:高浓度当归注射液对6-OHDA诱导PC12细胞帕金森病体外模型具有保护作用,当归注射液可能通过特定信号通路激活SIRT1、抑制6-OHDA诱导PC12细胞的凋亡而发挥其保护作用。
Objective:To investigate the effects of angelica injection on the expression of SIRT1 and on the neuronal injury of PC12 cells induced by 6-hydroxydopamine(6-OHDA).Methods:PC12 cells were treated with different concentrations of angelica injection(10,100,and 1 000 mg/L separately) two hours before the induction of 100 μmol/L 6-OHDA.After that,the PC12 cells was cultured for 24 hours and then MTT method was used to assay the cell's viability,the apoptosis was detected with flow cytometry,and the intensities of tyrosine hydroxylase(TH) and SIRT1 were detected with SABC immunocytochemistry staining.Results:Compared with 6-OHDA model group,the survival rate and TH positive cells in the 1 000 mg/L angelica injection group were increased(P0.01),and the number of SIRT1 positive cells were increased as well(P0.01).The rate of apoptosis in the 1 000 mg/L angelica injection groups was decreased significantly(P0.01).Conclusion:Angelica injection has neuroprotective effect on the 6-OHDA treated PC12 cell in vitro model of Parkinson's disease,probably through activating SIRT1 expression.
