血管内皮生长因子121在晶状体组织中的表达

Expression of Vascular Endothelial Growth Factor in Lens of Mice and Bovine

摘要 目的:探讨血管内皮生长因子121(VEGF 121)在小鼠囊膜组织和体外培养的新生小牛晶体上皮细胞中的表达情况。方法:分别完整的取下11个月龄的20只DBA/2J高眼压模型小鼠和20只正常BL6小鼠的晶状体囊膜组织,提取蛋白后,利用Western blot方法检测晶状体囊膜组织VEGF 121的表达。体外培养牛眼晶体上皮细胞,利用免疫细胞化学染色SP法检测晶体上皮细胞中VEGF121的表达。结果:Western blot方法显示两种小鼠囊膜均有VEGF121的表达,且两种小鼠的表达无明显差异。牛眼晶体上皮细胞也有VEGF121阳性颗粒的表达,阴性对照中未见阳性染色。结论:两种小鼠晶状体囊膜组织和体外培养的牛眼晶体上皮细胞均有VEGF 121的表达,表明晶状体中产生的VEGF 121可能参与小鼠胚胎发育和眼内某些新生血管性疾病的发生。 Objective:To study whether mouse lens capsule and cultured bovine lens epithelial cells express vascular endothelial growth factor 121(VEGF 121) in vitro or in vivo.Methods:The lens capsule of 20 DBA/2J intraocular hypertension model mice and 20 normal BL6 mice aged 11 months were taken off completely,and then their proteins were extracted,and the expression of VEGF 121 was detected with Western blot.Bovine lens epithelial cells were cultured in vitro and passaged three times,then the cells were transfered or cultured onto steriled cover glass,and the expression of VEGF 121 was detected with immunohistochemical technique-Supervision method.Results:VEGF 121 were significantly expressed both in the lens capsule of two kinds of mice with no difference and in the cultured bovine lens epithelial cells.No immunostaining was seen in the negative control.Conclusion:The presence of VEGF 121 in lens capsule of mice and cultured bovine lens epithelial cells were confirmed.It suggests that VEGF 121 may play an important role in the embryonic development and the diseases of neovascularization in the eyes.
