血性脑脊液对人胎脑神经干细胞影响的体外研究 被引量:1

Effect of Hemorrhagic Cerebrospinal Fluid on Human Embryonic Neural Stem Cells

摘要 目的:研究血性脑脊液对人胎脑神经干细胞增殖和分化的影响。方法:分别用人的血性脑脊液和正常脑脊液培养神经干细胞,分为试验组(血性脑脊液组)和对照组(正常脑脊液组),分别于不同时间观察神经干细胞在两组脑脊液中的生长及增殖分化情况,并应用免疫细胞化学技术对两组脑脊液中神经干细胞的分化进行鉴定和比较。结果:神经干细胞在血性脑脊液和正常脑脊液中均能存活,并且均能增殖和分化。神经干细胞在血性脑脊液中的贴壁分化速度快于正常脑脊液,分化比例也高。神经干细胞在血性脑脊液中倾向于向胶质细胞方向分化,在正常脑脊液中倾向于向神经元方向分化。结论:血性脑脊液对神经干细胞的迁移和分化产生了一定的影响。 Objective:To investigate the changes in growth,proliferation and differentiation of neural stem cells(NSCs) in human hemorrhagic cerebrospinal fluid(CSF).Methods:The NSCs were cultured in human hemorrhagic CSF from brain injury patients(hemorrhagic CSF group) or in CSF from normal subjects(normal CSF group).The growth,proliferation and differentiation pattern of NSCs in the two groups were observed at different time.The differentiated cells were identified by immunocytochemistry and their proportion was compared.Results:The NSCs could survive,proliferate and differentiate in hemorrhagic CSF and normal CSF.The attachment and differentiation speed of NSCs was faster,and the attachment ratio was higher in hemorrhagic CSF than in normal CSF.More NSCs were differentiated into astrocytes in hemorrhagic CSF,while more NSCs were differentiated into neurons in normal CSF.Conclusion:Hemorrhagic CSF has an impact on the proliferation and differentiation of human neural stem cells.
