GnRH Ⅱ与GnRH Ⅰ a对子宫内膜异位症患者离体间质细胞增殖抑制的影响 被引量:5

摘要 目的:探讨Ⅱ型促性腺激素释放激素(gonadotropin-releasing hormoneⅡ,GnRHⅡ)与Ⅰ型促性腺激素释放激素激动剂(gonadotropin-releasing hormoneⅠagonist,GnRHⅠa)对子宫内膜异位症(endometriosis,EMs)患者离体培养的子宫内膜间质细胞增殖抑制的影响。方法:将不同浓度的GnRHⅡ或GnRHⅠa加入在位及异位离体培养内膜间质细胞培养液中培养24,48及72 h,用MTT法测定间质细胞的存活情况,计算抑制率并进行比较。结果:GnRHⅡ或GnRHⅠa对离体培养的子宫内膜间质细胞的细胞增殖抑制率呈剂量和时间依赖性,差异均有统计学意义(P<0.05);且对异位子宫内膜间质细胞的增殖抑制率高于在位,差异有统计学意义(P<0.05)。GnRHⅡ对离体间质细胞的抑制作用强于GnRHⅠa,差异有统计学意义(P<0.05)。结论:GnRHⅡ对EMs患者离体培养子宫内膜间质细胞,尤其对异位子宫内膜间质细胞的增殖有明显的抑制作用,呈剂量和时间依赖性;且抑制作用明显强于GnRHⅠa,提示GnRHⅡ有望成为治疗子宫内膜异位症更有效的新药。 Objective To investigate the inhibitory effect of gonadotropin-releasing hormone Ⅱ(GnRHⅡ) and gonadotropin-releasing hormoneⅠagonist(GnRH Ⅰa) on the proliferation of endometrial stromal cells in vitro from endometriosis patients.Methods Different concentrations of GnRHⅡ or GnRH Ⅰa were added into the cultured endometrial stromal cells in vitro to detect the cell proliferation inhibition by MTT test.Results The inhibitory rate of GnRHⅡ or GnRH Ⅰa on eutopic and ectopic endometrial stromal cells in vitro was both dose-and time-dependent(P0.05).Effect of GnRHⅡ or GnRH Ⅰa on the inhibitory rate of ectopic endometrial stromal cells was significantly higher than that of eutopic(P0.05).GnRH Ⅱhad a higher inhibitory rate on the endometrial stromal cells in vitro than did GnRH Ⅰa(P0.05).Conclusion GnRHⅡ has more antiproliferative effect on endometrial stromal cells than GnRH Ⅰa in vitro,especially on ectopic endometrial stromal cells,suggesting that GnRHⅡ may be more effective than GnRH Ⅰa on endometriosis.
