摘要
利用混合悬浮培养的方法,构建了一个由重组大肠杆菌的谷胱甘肽生物合成酶系与面包酵母的糖酵解途径组成的ATP再生系统,验证了该系统用于谷胱甘肽(GSH)生物合成的可行性.研究了葡萄糖浓度、细胞量对耦合系统合成GSH 能力的影响,讨论了所构建的ATP再生系统中GSH 合成能力较低的可能原因.
An ATP regeneration system composed of the glutathione-synthetase in the recombinant Escherichia coli WSH KE1 and the EMP pathway in the Sacharomyces cerevisiae WSH J7 was established by the mixed culture of the two kinds of microorganisms in suspension. The feasibility of the system used in the biosynthesis of glutathione was verified. The effect of the glucose concentration and the biomass of the two kinds of microorganisms on the biosynthesis of glutathione was studied. The possible reason of the weaker ability of glutathione synthesis in the established ATP regeneration system was discussed.
