摘要
目的探讨正丁酸钠对人肝癌细胞恶性表型的逆转作用。方法以0.2mmol/L的正丁酸钠(sodiumbutyrate,SB)作用于培养的人肝癌细胞GHC-3,动态观察癌细胞生长、形态、ConA凝集性、琼脂集落形成率、染色体数目、DNA合成、DNA含量等表型的变化。结果SB作用7天,人肝癌细胞生长抑制;SB作用15天ConA凝集现象减弱;作用30天后细胞形态由多角型转变为棱形,细胞ConA凝集现象基本消失;经SB作用15天、30天、45天的细胞球脂集落形成率由对照组的4.54%下降为2.53%、1.45%、1.31%;SB作用30天、45天细胞3小时3H-TdR掺入率由2330.24士10.31cpm/105cells下降为1372.5123.41cpm/105cells和1136.23土17.23cpm/105cells,细胞DNA含量由198.511.18μg/107cells下降为134.78土0.53μg/107cells和104.78土0.53μg/107cells;均有显著差异(P<0.01)。染色体主流范围变窄,二倍体比例逐渐升高而亚三倍体比例逐渐下降,三倍体消失。结论人肝癌细胞在正了酸钠持续作用下,某些恶性表型可发生进行性逆转。
To observe the phenotypic changes of human hepatocarcinoma cell line induced by sodium butyrate. Methods GHC-3 cell line was cultured in vitro in sodiurn butyrate in a concentration of 0. 2mmol/L to observe the changes of cell morphology,growth rate, ConA aggluti nalbility, colony-forming rate, chromosome number, 3H-TdR incorporation rate, DNA concentration. Results Morphology changed from polygonal to spindle-shaped,proliferation rate was inhibited sig nificantly,ConA agglutinability disappeared. Colony-forming rate decreased gradually, the major range of chromosome turned from 52 - 59 to 44 - 45, the pencentages of diploid and hypodiploid increased gradually; the 3H-TdR incorporation rate and DNA concentration decreased gradually. Conclusion the malignant phenotypes of the human hepatocarcinoma cell line, GHC-3,can be reversed by sodium butyrate.
