高效液相色谱法同时测定血浆中的肾上腺素和去甲肾上腺素 被引量:8

摘要 为同时测定人血浆中肾上腺素和去甲肾上腺素浓度,建立了高效液相色谱-电化学检测方法,采用YWG-C18分析柱,3,4-二羟基苄胺作内标,以0.05m ol/L醋酸盐缓冲液(pH3.0)-甲醇-0.05m ol/LEDTA-Na2(78∶20∶2)为流动相,每1L流动相含SDS200m g,流速为1.0 m l/m in。去甲肾上腺素和肾上腺素及内标的保留时间分别为7.3、8.3 和10.4 分钟,线性范围均为31.25~4000ng/L(NE:r= 0.9999,E:r= 0.9996)。去甲肾上腺素和肾上腺素日内RSD分别低于1.87% 和2.07% ,日间RSD分别小于8.92% 和9.65% ,平均萃取回收率分别为93.5% 和91.3% 。提示本方法快速简便,灵敏准确,适用于嗜铬细胞瘤的鉴别诊断和临床研究。 A sensitive and rapid method was reported for the determination of nerepinephrine(NE) and epinephrine(E) in plasma by using high performance liquid chromatography with an electrochemical detector(HPLC EC) and YWG C 18 column(5u).3,4 Dihydroxybenzylamine(DHBA) was used as an internal standard. The mobile phase of 0.05mol/L actate buffer(pH3.0) methanol 0.05mol/L EDTA Na 2 (78∶20∶2) with of 200mg/L SDS solution was pumped through the column with a flow rate of 1.0ml/min. The lengths of retention time for NE, E and DHBA(IS) was 7.3 min, 8.3 min and 10.4min, respectively. The standard curve was linear within the concentration range of 31.25 to 4000ng/L for NE and E (NE: r =0.9999 E: r =0.9996), The within day RSD(%) were less than 1.87% for NE and less than 2.07% for E, inter day RSD(%) were less than 8.92% for NE and less than 9.65% for E respectively. The average extraction recovery(%) for NE and E were 93.5%±5.50% and 91.3%±1.77% respectively. This method has been applied to the diagnosis of pheochromocytoma and clinical research.
