葛根活性部位的纯化工艺研究

摘要 目的研究大孔吸附树脂纯化葛根活性部位的条件和参数。方法比较了4种大孔吸附树脂对葛根总异黄酮的吸附性能;以树脂吸附量和解吸率为指标,对大孔吸附树脂纯化葛根活性部位的工艺进行筛选。结果 AB-8大孔吸附树脂对葛根总异黄酮的吸附性能最好,其纯化的最佳条件为:原液浓度0.13 g生药/mL,流速为0.5 mL/min,pH值为原液pH值(4.5左右)。洗脱剂选择80%的乙醇溶液。洗脱流速为2mL/min。结论 AB-8大孔吸附树脂可用于葛根活性部位的纯化,纯化后活性部位的纯度达到70%以上。 Objective To study the conditions and parameters of purifying the effective position from Puerariae Radix. Methods Absorption capacity of four resins for pueraria total isoflavones was compared. The adsorption and desoption ability were carried out to select the best marcoporous resin. Results The AB-8 resin was the best of the four resins.The optimum process condition was 80% ethanol as eluting solvent,the flow rate at 0.5mL/min,pH at about 4.5,the solution concentration at 0.13g/mL. and the eluting velocity was 2mL/min. Conclusion The AB-8 resin could be used to purify the effective position of Pucrariae Radix. The purification was over 70%. The process is simple and convenient and the regeneration of resin is easy.
