丹参酮ⅡA对急性肾功能衰竭大鼠肾脏的保护作用 被引量:9

摘要 目的探讨丹参酮ⅡA对急性肾功能衰竭(AFR)大鼠肾小管上皮细胞及肾功能的作用。方法将64只大鼠分为4组:对照组、模型组、丹参酮低剂量组和丹参酮高剂量组,每组又分为48 h组和72 h组两个亚组。应用肌肉内注射甘油制备大鼠AFR模型。然后腹腔内注射不同浓度的丹参酮ⅡA进行干预。然后在48 h和72 h检测血尿素氮(BUN)、肌酐(Scr)和24 h尿蛋白定量;同时制作组织切片,观察肾组织结构改变,并应用免疫组织化学方法检测肾组织Ki-67的表达。结果丹参酮高剂量72 h组的各项病理损害指标均低于其他各组(P<0.05);丹参酮高剂量48 h及72 h组的Ki-67阳性细胞计数明显高于其他各组(P<0.05)。结论丹参酮ⅡA对急性损伤的肾小管上皮细胞具有减轻病变和促进再生的作用,可明显改善ARF大鼠的肾功能。 Objective To explore the protective effect of Tanshinone IIA on kidney in acute renal failure rats. Methods The 64 rats were divided into 4 groups,including control group, model group, Tanshinone low dose group and Tanshinone high dose group. According to observing time, the each group was further divided into 2 subgroups, 48h group and 72h group. The rats were injected glycerin saline to establish ARF animal model, then Tanshinone was injected with different dose. 48 hours and 72 hours later, BUN, Scr and urinary protein were detected. And tissue sections with HE staining and immunohistochemistry were made to observe the pathological changes and Ki-67 expressing. Result The pathological damage indexes in Tanshinone high dose group (72h) were significantly lower than that in other groups (P〈0.05) ;The Ki-67 positive cells count in Tanshinone high dose groups (48h and 72h) were significantly higher than that in other groups (P〈0. 05). Conclusion Tanshinone IIA can alleviate the lesion of tubular epithelial and promote their regeneration in ARF rats, by which improved the renal function of ARF rats.
