EDTA缓冲液在微波热修复神经组织糖皮质激素受体免疫组化染色中的应用

Application of EDTA Buffer Solution with Microwave Antigen Retrieval for Immunohistochemical Staining of Glucocorticoid Receptor in nerve tissue

目的比较不同修复液对大鼠糖皮质激素受体(GR)在神经染色效果的影响。方法分别采用枸椽酸修复液,pH8.5、9.0、9.5的EDTA修复液对大脑切片进行微波修复,比较不同的修复液显色阳性细胞数和OD值。结果 EDTA修复液显色效果更明显,pH9.0的EDTA修复液显色效果最明显。

Objective To detect the efficacy of different repair solution in immunohistochemical staining of glucocorticoid receptor （GR） in nerve tissue. Methods Two kinds of buffur solution of Sodium citrate and EDTA with pH 8. 5, 9.0, 9.5 with microwave antigen retrideve was used to stain GR in brain slice. The cell number and OD of GR in different retrievel solution were compared. Results The effect in EDTA was better than in sodium citrate, pH 9.0 was the best.
