摘要
目的:建立鸦胆子中3种苦木内酯的高效液相色谱含量测定方法。方法:采用phenomenex C18色谱柱(4.6 mm×150 mm,5μm),甲醇-水梯度洗脱,流速1.0 mL.min-1,检测波长270 nm。结果:鸦胆苦素D在2.52~12.60μg(r=0.9996),鸦胆苦醇在2.19~10.95μg(r=0.999 6),鸦胆因H在2.91~14.55μg(r=0.999 8)线性关系良好,鸦胆因D平均回收率100.01%,RSD 0.31%(n=6);鸦胆苦醇平均回收率100.95%,RSD 1.7%(n=6);鸦胆因H平均回收率100.43%,RSD1.7%(n=6)。结论:3批样品测定结果表明,该方法简便、准确,可用于鸦胆子中3种苦木内酯的含量测定。
Objective: To develop an HPLC method for quantitative determination of three quassinoids in Brucea javanic. Method: The determination was carried out on a phenomenex C18 column (4. 6 mm×250 mm,5 μm ) with gradient elution program of methol-water at a flow rate of 1.0 mL·min^-1, and the detection wavelength was 270 nm. Result: Linearites of bruceine D, brusatol and bruceine H were good ( r =0.999 6, 0.999 6, and 0.999 8) in ranges of 2.52-12.60, 2.19-10.95, and 2.91-14.55 μg, respectively. The average recoveries of bruceine D, brusatol and bruceine H were 100.01%, 100.95% and 100.43% respectively, and RSD of the above three compounds were 0.31% ( n =6), 1.7% ( n =6) and 1.7% ( n =6), respectively. Conclusion: The determination results of three batch samples showed that the method was simple, accurate and could be used in the quantitative determination of three components in the B. javanica.
