大承气汤对内毒素休克家兔白细胞内游离钙浓度的影响

EFFECT OF DANCHENGQI DICOCTION ON INTRACELLULAR FREE CALCIUM CONCENTRATION IN RABBIT SUFFERING FROM ENDOTOXIN SHOCK

目的：利用内毒素休克模型研究大承气汤对内毒素休克家兔白细胞内游离钙浓度的影响。方法：采用Ｆｕｒａ－２／ＡＭ 负载及荧光法测定对照组、内毒素休克组和大承气汤治疗组家兔白细胞内游离钙离子浓度。结果：与对照组比较，模型组的白细胞内钙离子浓度有明显升高（Ｐ＜ ０．０１）；与模型组比较，治疗组白细胞内钙离子浓度明显降低（Ｐ＜ ０．０１）。结论：大承气汤可使休克家兔白细胞内游离钙浓度明显降低，这是由于大承气汤抗钙超负荷，减轻其对组织的损伤，从而对细胞功能的调控起到保护作用。

Objective: To investigate effect of dachengqi dicoction on intracellular free Calcium concentrtion ([Ca 2+ ] i) in rabbit suffering from endotoxin shock using model of endotoxin shock. Mathods: Fura 2/AM load and fluorescent techniques were employed to demonstrate [Ca 2+ ] i of control group, endotoxin shock group and treatment group. Results: The value of [Ca 2+ ] i of endotoxin shock group is higher obviously compared with that of control group(p<0.01); the value of [Ca 2+ ] i of treatment group is lower obviously compared with that of shock group(p<0.01). Conclusion: Dachengqi dicoction may play an important role in reducing [Ca 2+ ] i of endotoxin shock of rabbit by inhibiting overload of Calcium ion ,it can reduce impairment of tissues and has a protective effect on regulation of cells function.
